Terraria can be best explained as a mix of the old-school 2-D side-scrolling exploration action games Metroid and Castlevania with a bit of the sandbox building/collection genre - made popular with games such as Minecraft - mixed in. In contrast to Minecraft's "build first, explore/fight later" gameplay style (the ratio might change a bit as Minecraft gets further in development), the focus of Terraria is first and foremost on exploration/loot/combat with building on the side. Like Minecraft, Terraria also has support for multiplayer. It's not a game heavy on the visuals, so modern graphics whores need not apply, but it has a nice style to it nonetheless which has plenty of appeal.
Unlike Minecraft, this game isn't about the individual world you live in (although it's nice to build fancy structures and what-not in worlds themselves). It's about the character you build and the swath of inventory options you have. Simply put, there are TONS of weapons, armors, and items to find and craft in Terraria, offering up lots of different playing styles and building options. You can set up farms and even craft potions (health, mana, buffs) via an alchemy system. One of the best things about Terraria is that you create a character, proceed to play the game to power him/her up with new weapons/armor/items, and can then take this supercharged character to other worlds and even into multiplayer. There are piggy bank items which act like a linked storage chest for your character which allows you to funnel items around a world and even between other worlds!
Terraria is still in heavy development, and actually was released early due to the insane demand for it. Thus, you can expect Terraria will have plenty of future updates adding even more content and loot to find.

Lots of crazy stuff. Building in 2-D (probably) doesn't have quite the same feel as building in 3-D like Minecraft, but it still has its charm.

The 1st of December brought an absolutely amazing Terraria update with it. This v1.1 update added over 220 new items, 30+ new mobs, 4 new bosses, an entire hardmode for worlds that is enabled only after beating a certain new boss (and thereby changing your existing world irreversibly), higher weapon/armor/item tiers, and lots of other tweaks/fixes. It was designed to be an expansion pack's worth of content.
I never put much time into this game after originally getting it because 1. I got lost with regards to what the hell I had to do, and 2. (IMO) the update to the game shortly after I bought it made the game frustrating as hell difficulty-wise midway through character development.
Anyways, after 5 months of not playing, this v1.1 update has rekindled my interest. Here is the trailer:
